An antibody specific for RNASE2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRNASE2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for RNASE2 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RNASE2 bound in the initial step. The color development is stopped and the intensity of the color is measured. The open reading frame encodes a 134-amino acid polypeptide with a molecµLar mass of 15.5 kD and a 27-residue N-terminal hydrophobic leader sequence. The sequence of the mature polypeptide was identical to that reported for urinary ribonuclease and to the N-terminal sequence of liver ribonuclease. Similarities to the ribonucleases of pancreas and angiogenin indicate that EDN belongs to the ribonuclease mµLtigene family. This gene is also symbolized RNS2 for ribonuclease 2.
