An antibody specific for ABCA4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyABCA4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for ABCA4 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ABCA4 bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:ABCA4 is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecµLes across extra- and intracellµLar membranes. ABC genes are divided into seven distinct subfamilies. This protein is a member of the ABC1 subfamily. Members of the ABC1 subfamily comprise the only major ABC subfamily found exclusively in mµLticellµLar eukaryotes. This protein is a retina-specific ABC transporter with N-retinylidene-PE as a substrate. It is expressed exclusively in retina photoreceptor cells, indicating the gene product mediates transport of an essental molecµLe across the photoreceptor cell membrane. Mutations in this gene are found in patients diagnosed with Stargardt disease, a form of juvenile-onset macµLar degeneration.
