An antibody specific for PRAMEF1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRAMEF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for PRAMEF1 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRAMEF1 bound in the initial step. The color development is stopped and the intensity of the color is measured. It is composed of repeating 20–30 amino acid stretches that are unusually rich in the hydrophobic amino acid leucine. Typically, each repeat unit has beta strand-turn-alpha helix structure, and the assembled domain, composed of many such repeats, has a horseshoe shape with an interior parallel beta sheet and an exterior array of helices. One face of the beta sheet and one side of the helix array are exposed to solvent and are therefore dominated by hydrophilic residues. The region between the helices and sheets is the protein's hydrophobic core and is tightly sterically packed with leucine residues.
